Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.13120G>A (p.Val4374Met), citing Ambry Variant Classification Scheme 2023: The p.V4374M variant (also known as c.13120G>A), located in coding exon 80 of the DNAH11 gene, results from a G to A substitution at nucleotide position 13120. The valine at codon 4374 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be and deleterious by PolyPhen and SIFT in silico analyses, respectively. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
