Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001289808.2(CRYAB):c.130C>A (p.Leu44Met), citing Ambry Variant Classification Scheme 2023: The p.L44M variant (also known as c.130C>A), located in coding exon 1 of the CRYAB gene, results from a C to A substitution at nucleotide position 130. The leucine at codon 44 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
