Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005477.3(HCN4):c.130A>T (p.Ser44Cys), citing Ambry Variant Classification Scheme 2023: The p.S44C variant (also known as c.130A>T), located in coding exon 1 of the HCN4 gene, results from an A to T substitution at nucleotide position 130. The serine at codon 44 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
