NM_000492.4(CFTR):c.1309G>T (p.Gly437Cys) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G437C variant (also known as c.1309G>T), located in coding exon 10 of the CFTR gene, results from a G to T substitution at nucleotide position 1309. The glycine at codon 437 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
