Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.13070G>A (p.Arg4357Gln), citing Ambry Variant Classification Scheme 2023: The p.R4357Q variant (also known as c.13070G>A), located in coding exon 80 of the DNAH11 gene, results from a G to A substitution at nucleotide position 13070. The arginine at codon 4357 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, glutamine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
