NM_001103.4(ACTN2):c.1300C>G (p.Leu434Val) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L434V variant (also known as c.1300C>G), located in coding exon 12 of the ACTN2 gene, results from a C to G substitution at nucleotide position 1300. The leucine at codon 434 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
