Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.12975G>C (p.Gln4325His), citing Ambry Variant Classification Scheme 2023: The p.Q4325H variant (also known as c.12975G>C), located in coding exon 75 of the DNAH5 gene, results from a G to C substitution at nucleotide position 12975. The glutamine at codon 4325 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
