Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.1017T>A (p.Asp339Glu), citing Ambry Variant Classification Scheme 2023: The p.D333E variant (also known as c.999T>A), located in coding exon 10 of the KIF1B gene, results from a T to A substitution at nucleotide position 999. The aspartic acid at codon 333 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
