NM_000020.3(ACVRL1):c.999C>A (p.Ser333Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S333R variant (also known as c.999C>A), located in coding exon 6 of the ACVRL1 gene, results from a C to A substitution at nucleotide position 999. The serine at codon 333 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
