NM_014845.6(FIG4):c.998C>T (p.Ser333Phe) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S333F variant (also known as c.998C>T), located in coding exon 9 of the FIG4 gene, results from a C to T substitution at nucleotide position 998. The serine at codon 333 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
