NM_003579.4(RAD54L):c.993T>G (p.Asp331Glu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D331E variant (also known as c.993T>G), located in coding exon 9 of the RAD54L gene, results from a T to G substitution at nucleotide position 993. The aspartic acid at codon 331 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
