NM_006231.4(POLE):c.991C>G (p.Pro331Ala) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P331A variant (also known as c.991C>G), located in coding exon 10 of the POLE gene, results from a C to G substitution at nucleotide position 991. The proline at codon 331 is replaced by alanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
