Uncertain significance — the classification assigned by Ambry Genetics to NM_004517.4(ILK):c.98A>C (p.His33Pro), citing Ambry Variant Classification Scheme 2023: The p.H33P variant (also known as c.98A>C), located in coding exon 2 of the ILK gene, results from an A to C substitution at nucleotide position 98. The histidine at codon 33 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
