Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_005585.5(SMAD6):c.1283G>A (p.Arg428His), citing Ambry Variant Classification Scheme 2023: The p.R428H variant (also known as c.1283G>A), located in coding exon 4 of the SMAD6 gene, results from a G to A substitution at nucleotide position 1283. The arginine at codon 428 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
