NM_000492.4(CFTR):c.1282C>A (p.Leu428Ile) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L428I variant (also known as c.1282C>A), located in coding exon 10 of the CFTR gene, results from a C to A substitution at nucleotide position 1282. The leucine at codon 428 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, isoleucine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
