Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.977C>G (p.Ala326Gly), citing Ambry Variant Classification Scheme 2023: The p.A326G variant (also known as c.977C>G), located in coding exon 8 of the CFTR gene, results from a C to G substitution at nucleotide position 977. The alanine at codon 326 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
