NM_001378454.1(ALMS1):c.9757G>C (p.Val3253Leu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V3254L variant (also known as c.9760G>C), located in coding exon 11 of the ALMS1 gene, results from a G to C substitution at nucleotide position 9760. The valine at codon 3254 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
