Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001211.6(BUB1B):c.971G>T (p.Arg324Leu), citing Ambry Variant Classification Scheme 2023: The p.R324L variant (also known as c.971G>T), located in coding exon 8 of the BUB1B gene, results from a G to T substitution at nucleotide position 971. The arginine at codon 324 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
