NM_000314.8(PTEN):c.969T>A (p.Asn323Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N323K variant (also known as c.969T>A), located in coding exon 8 of the PTEN gene, results from a T to A substitution at nucleotide position 969. The asparagine at codon 323 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. This missense alteration is located in a region that has a low rate of benign missense variation (Lek M et al.
