Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001903.5(CTNNA1):c.963C>G (p.Asp321Glu), citing Ambry Variant Classification Scheme 2023: The p.D321E variant (also known as c.963C>G), located in coding exon 6 of the CTNNA1 gene, results from a C to G substitution at nucleotide position 963. The aspartic acid at codon 321 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
