NM_000268.4(NF2):c.960G>C (p.Gln320His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q320H variant (also known as c.960G>C), located in coding exon 10 of the NF2 gene, results from a G to C substitution at nucleotide position 960. The glutamine at codon 320 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
