Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003977.4(AIP):c.95C>A (p.Thr32Asn), citing Ambry Variant Classification Scheme 2023: The p.T32N variant (also known as c.95C>A), located in coding exon 1 of the AIP gene, results from a C to A substitution at nucleotide position 95. The threonine at codon 32 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
