Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_015450.3(POT1):c.950G>A (p.Ser317Asn), citing Ambry Variant Classification Scheme 2023: The p.S317N variant (also known as c.950G>A) is located in coding exon 8 of the POT1 gene. The serine at codon 317 is replaced by asparagine, an amino acid with highly similar properties. This change occurs in the first base pair of coding exon 8. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
