NM_001148.6(ANK2):c.9422A>G (p.Glu3141Gly) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E3141G variant (also known as c.9422A>G), located in coding exon 38 of the ANK2 gene, results from an A to G substitution at nucleotide position 9422. This exon is expressed solely in brain (Mohler PJ et al. Circulation. 2007;115(4):432-41). The glutamic acid at codon 3141 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
