NM_001042492.3(NF1):c.940A>G (p.Arg314Gly) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R314G variant (also known as c.940A>G), located in coding exon 9 of the NF1 gene, results from an A to G substitution at nucleotide position 940. The arginine at codon 314 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
