Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.930C>A (p.Tyr310Ter), citing Ambry Variant Classification Scheme 2023: The p.Y310* pathogenic mutation (also known as c.930C>A), located in coding exon 9 of the PMS2 gene, results from a C to A substitution at nucleotide position 930. This changes the amino acid from a tyrosine to a stop codon within coding exon 9. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
