Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.9256G>C (p.Asp3086His), citing Ambry Variant Classification Scheme 2023: The p.D3086H variant (also known as c.9256G>C), located in coding exon 38 of the AKAP9 gene, results from a G to C substitution at nucleotide position 9256. The aspartic acid at codon 3086 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
