Likely pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000455.5(STK11):c.920+2_920+31del, citing Ambry Variant Classification Scheme 2023: The c.920+2_920+31del30 intronic variant, located in intron 7 of the STK11 gene, results from a deletion of 30 nucleotides within intron 7 of the STK11 gene. This nucleotide region is somewhat conserved in available vertebrate species. Using the BDGP and ESEfinder splice site prediction tools, this alteration is predicted to abolish the native splice donor site; however, direct evidence is unavailable. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as likely pathogenic.
