Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000314.4(PTEN):c.-917G>T, citing Ambry Variant Classification Scheme 2023. This variant lies in the PTEN gene (transcript NM_000314.4) at 917 bases upstream of the translation start (5' untranslated region), where G is replaced by T. Submitter rationale: The c.-917G>T variant is located in the 5' untranslated region (5’ UTR) of the PTEN gene. This variant results from a G to T substitution 917 bases upstream from the first translated codon. This variant is located in the promoter region of the PTEN gene, but its potential impact on PTEN regulation has not yet been investigated (Zhou XP et al. Am J Hum Genet. 2003;73:404-411). Allele frequency data for this nucleotide position is not currently available from the NHLBI Exome Sequencing Project (ESP) or the 1000 Genomes Project and the alteration is not currently listed in the Database of Single Nucleotide Polymorphisms (dbSNP). This nucleotide position is not well conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of c.-917G>T remains unclear.
