NM_000492.4(CFTR):c.911A>C (p.Tyr304Ser) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y304S variant (also known as c.911A>C), located in coding exon 8 of the CFTR gene, results from an A to C substitution at nucleotide position 911. The tyrosine at codon 304 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
