NM_001148.6(ANK2):c.9117T>G (p.Asp3039Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D3039E variant (also known as c.9117T>G), located in coding exon 38 of the ANK2 gene, results from a T to G substitution at nucleotide position 9117. This exon is expressed solely in brain (Mohler PJ et al. Circulation. 2007;115(4):432-41). The aspartic acid at codon 3039 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, glutamic acid is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
