NM_000492.4(CFTR):c.901T>C (p.Tyr301His) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y301H variant (also known as c.901T>C), located in coding exon 8 of the CFTR gene, results from a T to C substitution at nucleotide position 901. The tyrosine at codon 301 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
