NM_003242.6(TGFBR2):c.901C>T (p.His301Tyr) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H301Y variant (also known as c.901C>T), located in coding exon 4 of the TGFBR2 gene, results from a C to T substitution at nucleotide position 901. The histidine at codon 301 is replaced by tyrosine, an amino acid with similar properties, and is located in a serine/threonine protein kinase domain. A different variant affecting this codon (p.H301R, c.902A>G) was reported in 3 individuals from a Loeys-Dietz sydrome cohort; however, clinical details were not provided (Frischmeyer-Guerrerio PA et al. Sci Transl Med, 2013 Jul;5:195ra94). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
