This variant lies in the APOC3 gene (transcript NM_000040.3) at coding-DNA position 90 through coding-DNA position 95, duplicating 6 bases. Submitter rationale: The c.90_95dupCTTCAT variant (also known as p.F31_M32insIF), located in coding exon 2 of the APOC3 gene, results from an in-frame duplication of CTTCAT at nucleotide positions 90 to 95. This results in the insertion of 2 extra residues (isoleucine and phenylalanine) between codons 31 and 32. This amino acid region is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
