Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.890A>T (p.His297Leu), citing Ambry Variant Classification Scheme 2023: The p.H297L variant (also known as c.890A>T), located in coding exon 9 of the BUB1 gene, results from an A to T substitution at nucleotide position 890. The histidine at codon 297 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
