NM_014391.3(ANKRD1):c.88G>T (p.Asp30Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D30Y variant (also known as c.88G>T), located in coding exon 2 of the ANKRD1 gene, results from a G to T substitution at nucleotide position 88. The aspartic acid at codon 30 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
