Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.905A>G (p.Asp302Gly), citing Ambry Variant Classification Scheme 2023: The p.D296G variant (also known as c.887A>G), located in coding exon 9 of the KIF1B gene, results from an A to G substitution at nucleotide position 887. The aspartic acid at codon 296 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
