Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.883-4T>G, citing Ambry Variant Classification Scheme 2023: The c.883-4T>G intronic variant results from a T to G substitution 4 nucleotides upstream from coding exon 5 in the DNAH11 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will not have any significant effect on splicing. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
