NM_001378454.1(ALMS1):c.8788T>C (p.Phe2930Leu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F2931L variant (also known as c.8791T>C), located in coding exon 10 of the ALMS1 gene, results from a T to C substitution at nucleotide position 8791. The phenylalanine at codon 2931 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
