Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.872C>T (p.Pro291Leu), citing Ambry Variant Classification Scheme 2023: The p.P291L variant (also known as c.872C>T), located in coding exon 10 of the NPAT gene, results from a C to T substitution at nucleotide position 872. The proline at codon 291 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
