NM_003072.5(SMARCA4):c.86G>T (p.Gly29Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G29V variant (also known as c.86G>T), located in coding exon 1 of the SMARCA4 gene, results from a G to T substitution at nucleotide position 86. The glycine at codon 29 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
