This variant lies in the DNAH5 gene (transcript NM_001369.3) at coding-DNA position 8690, where A is replaced by G; at the protein level this means replaces tyrosine at residue 2897 with cysteine — a missense variant. Submitter rationale: The p.Y2897C variant (also known as c.8690A>G), located in coding exon 52 of the DNAH5 gene, results from an A to G substitution at nucleotide position 8690. The tyrosine at codon 2897 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species.
