Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_181486.4(TBX5):c.868C>G (p.Gln290Glu), citing Ambry Variant Classification Scheme 2023: The p.Q290E variant (also known as c.868C>G), located in coding exon 7 of the TBX5 gene, results from a C to G substitution at nucleotide position 868. The glutamine at codon 290 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
