NM_006096.4(NDRG1):c.863A>C (p.Asp288Ala) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D288A variant (also known as c.863A>C), located in coding exon 13 of the NDRG1 gene, results from an A to C substitution at nucleotide position 863. The aspartic acid at codon 288 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
