Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002528.7(NTHL1):c.838C>T (p.Gln280Ter), citing Ambry Variant Classification Scheme 2023: The p.Q288* variant (also known as c.862C>T), located in coding exon 6 of the NTHL1 gene, results from a C to T substitution at nucleotide position 862. This changes the amino acid from a glutamine to a stop codon within coding exon 6. This alteration occurs at the 3' terminus of theNTHL1 gene, is not expected to trigger nonsense-mediated mRNAdecay, and only impacts the last 8% of the protein. The exact functional effect of this alteration is unknown. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
