Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.854A>C (p.Asp285Ala), citing Ambry Variant Classification Scheme 2023: The p.D285A variant (also known as c.854A>C), located in coding exon 10 of the NPAT gene, results from an A to C substitution at nucleotide position 854. The aspartic acid at codon 285 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
