Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.84G>T (p.Glu28Asp), citing Ambry Variant Classification Scheme 2023: The p.E28D variant (also known as c.84G>T), located in coding exon 2 of the ATM gene, results from a G to T substitution at nucleotide position 84. The glutamic acid at codon 28 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
