Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.8433_8632+531dup, citing Ambry Variant Classification Scheme 2023: The EX19dup gross duplication spans coding exon 19 in the BRCA2 gene. A duplication of this region, referred to as dup exon 20, was observed in two breast cancer families from Spain that had previously tested negative on BRCA1/2 sequencing (Gutierrez-Enriquez S et al. Breast Cancer Res Treat. 2007 May;103(1):103-7). Additional analysis to determine breakpoints identified that this duplication is in tandem and is predicted to result in a premature truncation causing a translational frameshift with a predicted alternate stop codon (Ambry internal data). As such, this alteration is interpreted as a disease-causing mutation.
