NM_001367721.1(CASK):c.838G>T (p.Asp280Tyr) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D280Y variant (also known as c.838G>T), located in coding exon 9 of the CASK gene, results from a G to T substitution at nucleotide position 838. The aspartic acid at codon 280 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
