Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_015141.4(GPD1L):c.837C>G (p.Phe279Leu), citing Ambry Variant Classification Scheme 2023: The p.F279L variant (also known as c.837C>G), located in coding exon 6 of the GPD1L gene, results from a C to G substitution at nucleotide position 837. The phenylalanine at codon 279 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
